It seems that things are really starting to pick up pace for the Captain America sequel, titled Captain America: The Winter Soldier. Many of the cast members have been put in place and the storyline is pretty much set in stone and now the finally characters are getting cast.

It was recently announced that Mary Elizabeth Winstead was the favourite to land the female lead role. Mary would play Sharon Carter (aka Agent 13), who is the niece of Peggy Carter, who was the main female character in the first Captain America movie.
Now it has been claimed that Lost actor Josh Holloway is the favourite to play one of the very physical male roles, which Marvel have already spoken about. These male roles are believed to be that of a villain, although that has yet to be confirmed, and Josh Holloway is expected to appear in Captain America: The Winter Soldier as one of these physical males.

While all of this news is well and good, nothing has yet been officially confirmed and there is still plenty of time before production begins for things to change. Captain America: The Winter Soldier is set to begin production in March 2013 and is expected to be released on April 4, 2014.
